二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

 申请人近年来一直致力于中枢免疫信号激活引起的神经元和突触的发育及功能障碍的机制研究。在模式识别受体的中枢功能研究中,发现并解析了发育关键期星形胶质细胞TLR4通路激活调控突触发育的新机制,这一工作于2016年以第一作者论文发表在《Journal of Cell Biology》(IF 9.362)上。随后同期杂志的评论称赞我们的发现为研究和治疗癫痫打开了全新之门。此外,申请人所在团队首次发现了重要的免疫信号分子脂筏结构中的磷脂酰肌醇-4,5-二磷酸在阿尔兹海默症早期纳摩尔级寡聚引起的突触前功能紊乱中起重要作用,选择性降低这些靶点的功能可以有效挽救的突触前效应及模型小鼠的认知功能。这一研究成果于2019年发表在国际著名期刊《Nature CommunicationsIF 13.811),申请人作为共同通讯作者。

申请人自201112�加入浙江大学神经科学研究所周煜东教授课题组以来,主要开展中枢神经系统(CNS)免疫信号调控神经元和突触发育及功能的机制研究。主持国家自然科学基金项目3项、浙江省自然科学基金项目1项,参与多个课题研究,先后发表研究论文10余篇,其中6篇为第一作者或通讯作者,包括Journal of Cell BiologyNature CommunicationsJournal of Neuroscience等国际著名期刊。

长期以来,CNS被认为具有免疫特权(immune-privileged),即正常的脑一般不会被外周免疫细胞侵入,其对环境的监控和抵抗由脑内的特定细胞(如小胶质细胞)来执行;只有在一些病理状态下,特别是血脑屏障遭受破坏的情况下,外周免疫细胞才能进入CNS。因此,传统上把CNS视为免疫过程并不活跃的区域。但近年来研究表明,CNS存在着非常活跃的先天免疫系统,许多原本在外周免疫系统中被发现的重要免疫分子及其信号通路也参与调控CNS功能。CNS免疫应答由能够识别与宿主不同的病原相关分子模式(PAMPs)以及宿主来源的损伤相关分子模式(DAMPs)的模式识别受体(PRRs)启动,例如主要识别PAMPsToll样受体(TLRs)。TLRs在小胶质细胞、星形胶质细胞和神经元都有表达,在先天性免疫系统中起关键作用,激活后可分泌一系列在生理情况下较低水平的促炎性细胞因子,如IL-1βIL-6TNF-α等。这些免疫细胞和免疫分子及细胞因子不仅参与了许多神经精神疾病的病理过程,还对神经元和突触的发生、神经元环路的形成、突触可塑性的调控等正常生理过程起了关键的调节作用。因此研究先天免疫系统中重要信号分子对CNS功能调控的机制不仅能在理论上更好地理解免疫系统与CNS的相互作用,还可以为多种神经精神疾病的病理研究和临床诊治提供新的线索。

近年来申请人着重开展了CNS免疫信号激活造成的神经元和突触发育及功能障碍的研究,做了不少原创性的工作,并发表有较高水平的文章。申请人发现并解析了发育关键期星形胶质细胞TLR4通路激活调控突触发育的新机制。除了PAMP及其信号途径对CNS的影响,申请人也开展了一些由非病原微生物感染而产生的DAMPCNS功能调控的研究。例如,长链饱和脂肪酸(LCSFA)是TLR4的配体,我们发现LCSFA与其受体结合后可激活丘脑室旁核(PVT)兴奋性神经元,造成动物的强迫进食行为。此外,我们发现重要的免疫信号分子——脂筏(lipid raft)结构中的主要成分磷脂酰肌醇-4,5-二磷酸(PIP2)参与寡聚引起的突触前功能紊乱。下面详细介绍申请人在神经免疫方面所取得的重要研究进展。

1. PAMP激活TLR调控中枢连接

星形胶质细胞是CNS内广泛分布的一类胶质细胞,参与多种生理和病理过程。在大脑发育过程中,星形胶质细胞通过调节轴突导向、突触形成、突触删除、突触功能、神经元存活等来协调脑内神经元环路的发育。星形胶质细胞的发育和功能异常与多种CNS疾病的发生,如癫痫、自闭症、努南综合征等相关。然而,星形胶质细胞如何在病理条件下调节神经元之间的突触联系目前并不清楚。另外,作为脑内重要的免疫细胞,星形胶质细胞TLR激活是否影响神经元之间的连接也是目前胶质细胞生物学和发育神经生物学研究所面临的重要科学问题。

我们发现相较于成年小鼠,外周PAMP刺激(细菌细胞壁成分脂多糖,LPS)对出生后2周的幼年小鼠的癫痫易感性具有较高的易化作用,并且这一易化作用与外周免疫刺激造成的海马锥体神经元树突分支增多、树突棘密度增高相关。外周PAMP刺激造成的幼年小鼠海马神经元兴奋性突触增多现象与星形胶质细胞的激活密切相关。利用海马神经元和星形胶质细胞共培养技术以及小鼠胚胎电转技术(IUE),我们进一步分析了幼年小鼠星形胶质细胞的免疫激活信号,发现星形胶质细胞TLR4-MyD88-ERK1/2通路的激活对兴奋性突触的发生有直接的促进作用,而抑制这一信号通路则可以挽救免疫刺激所造成的幼年小鼠的癫痫易感性。这些结果表明,在特定发育阶段星形胶质细胞中的免疫信号通路激活对兴奋性突触的形成具有正向调控作用,从而造成海马神经环路兴奋与抑制的失衡。该研究不仅有助于深入理解星形胶质细胞调控神经元环路发育的机制,而且为相关神经发育疾病的诊断和治疗提供了新思路。值得一提的是,所有工作完全在国内完成,并于201611�份在国际著名期刊《Journal of Cell Biology》上发表,申请人为第一作者。随后德国波恩大学的Christian Henneberger教授和英国伦敦大学学院的Christian Steinhäuser教授在同期对我们的论文发表了评论,称赞我们的发现为研究和治疗癫痫打开了全新之门。

2. 免疫信号分子在阿尔兹海默症(AD)中的新作用

脂筏结构在免疫细胞信号转导过程中起了很关键的作用,也是许多病毒侵入细胞的重要位点。脂筏结构中的主要成分PIP2含量的高低与脂筏中TLR的激活成正相关。免疫细胞脂筏中这些参与免疫信号转导的分子也在神经元脂筏中发挥着重要的作用。最近的研究表明,神经元脂筏中细胞型朊蛋白(PrPC)和代谢型谷氨酸受体5亚型(mGluR5)是寡聚的受体,寡聚与它们结合后可以使mGluR5及脂筏中其它信号分子(如蛋白激酶Fyn)激活。而mGluR5的激活显然可以改变细胞膜PIP2的含量,进一步影响神经细胞的生理功能。

我们最近的研究发现,引起的突触前功能紊乱(比如神经递质释放几率的下降)往往先于造成的突触后功能异常。在7�龄APP/PS1小鼠中首先被观察到的也是递质释放几率的降低。进一步研究表明,纳摩尔级的寡聚通过与突触前的PrPC-mGluR5结合,激活磷脂酶CPLC),引起突触前膜PIP2的含量降低,导致突触前谷氨酸释放几率的下降,从而引起突触功能损害。利用海马神经元微岛培养技术结合自突触(autapse)的电生理记录,我们发现胞内加入外源PIP2可以抵抗纳摩尔级的寡聚引起的谷氨酸释放几率的降低。采用cre-loxp技术选择性降低APP/PS1小鼠突触前mGluR5对配体的敏感程度,能够恢复APP/PS1小鼠的谷氨酸释放几率并改善小鼠的认知功能。以上结果表明:突触前mGluR5-PLCAD早期较低病理浓度的寡聚作用的主要靶点,针对这些靶点的拮抗剂可以有效地挽救的突触前效应。这一研究成果于2019年发表在国际著名期刊《Nature Communications》上,申请人为共同通讯作者。

3. DAMP激活TLR调控中枢功能

LCSFA是一典型的DAMP,可以被TLR4所识别。LCSFA可随血液循环进入脑内,因此研究LCSFA对能量代谢和进食行为的内稳态调控机制研究具有十分重要的理论意义和应用价值。目前这一领域的工作主要集中在研究LCSFA如何作用于下丘脑这个全身能量稳态调控的中枢来调节能量代谢和进食行为,而对进食的奖赏动机及其调控机制的关注相对较少。由异常进食动机驱动的强迫进食行为,即个体明知面临潜在的负性后果却仍无法抑制进食的行为,是肥胖病人持续过度进食的主要因素之一,严重影响肥胖的治疗,危害国民的身心健康。

我们发现高脂食物喂养或脑室注射棕榈酸(PA)可以诱发小鼠对美味食物的强迫性进食行为,提示进入CNSLCSFA可以改变动物的进食动机。高脂喂养或脑室注射PATLR4敲除小鼠的进食动机则没有影响,表明PA通过TLR4受体提高了小鼠对美味食物的进食动机。我们随后发现,PA可直接增加位于PVT的谷氨酸能神经元的兴奋性。用光遗传学手段特异性激活PVT兴奋性神经元可以诱发正常喂养小鼠的强迫性进食;而特异性抑制PVT兴奋性神经元活动则可减弱LCSFA诱发的强迫性进食。为了明确LCSFA介导强迫性进食行为的神经环路,我们利用AAV-ChR2顺行追踪病毒和RV-EnvA-DG/AAV helper逆行追踪病毒技术,结合光遗传方法,发现特异性激活PVT-下丘脑腹内侧核(VMH)兴奋性神经元通路可以诱发小鼠明显的强迫性进食行为。这些结果表明,LCSFA可以通过激活PVT-VMH通路增强小鼠的进食动机。我们的工作首次明确了除下丘脑之外,丘脑PVT神经元也起着监控饱和脂肪酸的作用。在食物匮乏的野生环境中,这一监控机制可保证动物持续的获取高能量食物。而在食物丰盛的现代社会,这一机制却造成了过多的能量摄入。这一部分工作已经完成,正在投稿中,申请人为共同通讯作者。


三、岗位工作思路&= #21450;预期目标

申请人对于岗位工作的自我定位是:科研、教学、公共服务平稳均衡的发展。在科研、教学、社会服务等方面岗位工作计划与预期目标如下:

1. 学术方面将开展以下3个研究方向:

1)免疫信号分子在阿尔兹海默病中的新作用。开展星形胶质细胞mGluR5在阿尔兹海默病早期对突触功能的调控及机制研究;

2)病原相关分子模式(PAMPs)激活TLR调控中枢连接。继续开展星形胶质细胞释放因子三螺旋重复胶原蛋白1Cthrc1)调控兴奋性突触发生的机制研究;

3)损伤相关分子模式(DAMPs)激活小胶质细胞调控中枢功能:解析高脂食物激活丘脑室旁核小胶质细胞引起的炎症反应介导小鼠强迫性进食行为。

完成相关的已投稿或待投稿的论文,包括:
