二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

1.环境内分泌干扰物的促肿瘤分子机制研究:(1)发现苯并芘可以上调肺癌细胞中长链非编码RNA-Linc00673的表达促进肺癌的进展,并首次阐明Linc00673作为miR-150的分子海绵通过上调miR-150的靶基因ZEB1促进肺癌上皮间质转化的分子机制,为肺癌的防治提供了新的思路及分子靶标;(2)发现氯氰菊酯在短时暴露条件下可通过miR-155/BCL6通路调控巨噬细胞的极化促进肺癌的发展,而在长时暴露条件下则可通过ROS介导的JNK/ERK信号通路促进巨噬细胞的凋亡,为氯氰菊酯对肿瘤相关巨噬细胞的毒理效应提供了新的理论依据;

2.肿瘤防治靶标的筛选研究:发现阿司匹林治疗肿瘤的新型时间窗及两类靶向有效人群(PTGS2阳性和PIK3CA突变型),为阿司匹林的肿瘤个体化防治方案提供了新的参考。

申请人的主要研究方向为分子毒理学及分子流行病学,主要学术成绩,贡献及科学价值包括以下两方面:

 环境内分泌干扰物的促肿瘤机制研究:

【申请人以重要的三类环境内分泌干扰物(芳香烃受体配体,雌激素受体配体,PPAR受体配体)为研究对象,研究其促肿瘤的细胞分子机制以及在人群中的致瘤效应】

1)发现了重要的环境内分泌干扰物苯并芘(大气污染主成分,芳香烃受体配体)可以通过上调肺癌细胞中长链非编码RNA-Linc00673的表达促进肺癌的进展,并进一步阐明了Linc00673可以作为miR-150的分子海绵上调miR150关键靶基因ZEB1从而促进肺癌细胞上皮间质转化的分子机制,在动物和人群模型中验证了Linc00673作为良好的肺癌分子干预靶标的潜能,前期相关工作已经发表在肿瘤JCR一区杂志Mol Cancer.2017.16(1): 118上(IF:6.2),并以此为基础获得国家自然基金支持,后续工作在投,该系列工作的完成不仅揭示了环境内分泌干扰物通过非编码RNA调控肿瘤进展的全新分子机制,也提供了潜在的肿瘤分子干预靶标。

2)发现了重要的环境内分泌干扰物氯氰菊酯(农药主成分之一,类雌激素样化合物)在短时暴露条件下通过miR155/BCL6途径促进肿瘤相关巨噬细胞从M1型向M2型转换从而促进肺癌的发展,该部分工作目前在投毒理学顶尖杂志Toxicological Sciences(under review);而在长时暴露条件下氯氰菊酯可以诱导巨噬细胞G1期阻滞并通过ROS介导的JNK/ERK信号直接导致巨噬细胞的凋亡,该部分相关工作发表在Int J Mol Sci.2016.Jun 17;17(6)该系列工作为氯氰菊酯对肿瘤相关巨噬细胞的潜在毒理学效应提供了新的依据

3)通过人群数据库分析重要的环境内分泌干扰物二噁英(大气污染主成分,芳香烃受体配体)内外暴露对人群全肿瘤谱发病率,死亡率的影响,完成量效分析,并筛选到与二噁英内外暴露均显著相关的肿瘤非霍奇金淋巴瘤,该部分工作发表在Sci Rep.2016.6: 38012上,为后续探索二噁英更精准的毒性效应人群及潜在的分子机制提供了全新的思路。

4)通过人群数据库分析发现重要的环境内分泌干扰物邻苯二甲酸酯类(塑化剂主成分,PPAR受体配体)及其代谢物的内暴露对女性乳腺癌及生殖系统肿瘤发病率的影响,并通过毒理生信数据库CTD(The comparative Toxicogenomics Database)鉴定出可能与邻苯二甲酸二辛酯(DEHP)促进乳腺癌发病密切相关的关键信号通路,包括P53依赖的凋亡信号通路,PPARa通路以及ATM激酶信号通路,该研究目前已被毒理学优秀杂志Reproductive Toxicology发表,为邻苯二甲酸酯类的毒性效应人群分类及其潜在的分子机制提供了理论依据。

 

 基于机制及人群研究的肿瘤防治靶标的筛选:

申请人以肿瘤分子机制研究为基础,结合流行病学与生物信息学技术,探索肿瘤防治的精准靶标】
1) 基于人群数据库分析发现了阿司匹林治疗肿瘤的新型时间窗,即只有在结直肠癌诊断之后使用阿司匹林才能改善结直肠癌患者的生存率(提前使用没有改善效果),为学术界对阿司匹林抗肿瘤机制的研究提供了新的思路,并筛选到2类阿司匹林的靶向有效人群(PTGS2阳性及PIK3CA突变型患者),该工作以Original Article的形式发表在国际消化道顶尖杂志Gut.2015.Sep;64(9):1419-25 (IF:16.7)),两年内被正面他引33次,被Nature reviews cancer、JCO、JNCIGastroenterology和Gut等一系列领域重要综述及研究所引用,为结直肠癌的个体化治疗以及阿司匹林进入肿瘤防治指南提供了新的依据,并获得2015年浙江大学医学院科研重大贡献奖;同时,申请人对微量元素摄入(抗氧化维生素,微量金属元素锌等)防控消化道肿瘤的效果做了系统,深入的系列研究,发表了包括International Journal of Cancer European Journal of NutrtionClinical Nutrition等领域TOP期刊5篇。

2) 以肿瘤模式细胞为筛选对象,通过一系列药物筛选和分子功能实验发现了和自噬拮抗剂氯喹联用可以大幅增强杀伤肿瘤细胞效果的新型单体化合物Physapubescin B,发表在氧化应激领域JCR一区顶尖杂志Redox Biology.2017.12: 198-207(5-year IF:6.9),为通过自噬和凋亡的细胞死亡途径筛选新的抗肿瘤分子靶标提供了参考。

3)以自建结直肠癌患者小样本人群队列,及2个西方独立队列(分别由国际著名分子流行病学专家C.C.HarrisKulda教授各自主持)的患者为研究对象,鉴定了血清miR-21可作为结直肠癌分子诊断及预后标记物的功能,相关论文发表在AACR肿瘤权威杂志Cancer Epidemiology Biomarkers&Prevention.2014.Dec;23(12):2783-92,该研究为肿瘤新型分子诊治靶标的筛选提供了新的依据。


三、岗位工作思路&= #21450;预期目标

下一阶段科研规划思路及目标:

1.继续开展对重要的环境内分泌干扰物(包括①雌激素受体配体BPA及其替代品BPS;②芳香烃受体配体苯并芘,二噁英;③PPAR受体配体DEHP及其代谢物MEHP)的致癌机制研究,以上述三种重要的环境内分泌干扰物为研究对象,以新一代组学及生物信息学技术为研究基础,探索环境内分泌干扰物对肿瘤细胞及肿瘤微环境(肿瘤相关巨噬细胞,肿瘤相关成纤维细胞,肿瘤相关血小板等)的综合调控机制,为分子毒理学研究开辟新的研究方向,并以机制研究为基础尝试寻找目标肿瘤的防治靶点;在机制研究过程中,依托于新加坡国立大学的研究基础以及已获得的国家,省自然基金的项目基础,重点筛选长链非编码RNA及自噬相关基因(ATG)在肿瘤进展中的功能研究,争取在下一阶段针对环境内分泌干扰物的肿瘤调控机制形成一个有系列性且亮点突出的学科方向,争取3年内发表1-2篇10分以上代表性工作以及5篇5分以上领域顶尖工作,并力争获得国家优秀青年基金立项。

2.以肿瘤分子机制研究为基础,开展肿瘤分子靶向药物筛选,以公卫大环境为导向,积极研究新型肿瘤防治药物(重点建库筛选可以拮抗肿瘤微环境的非肿瘤药物,比如抗血小板药物,抗糖尿病药物等;同时,和药学专家合作设计基于机制研究的靶向小分子药物),目前已经有2个具备良好潜力的对象完成了表型实验,力争和浙大附属医院等合作单位通力合作,为医学院及公卫学院创建一个有影响力的方向。

 

 下一阶段教学、科研带教、学科建设、社会工作等方面的工作任务和目标:

1.教学方面:参加公共卫生系教育改革,作为B角配合新加坡国立大学教授沈汉明(医学院校友,公共卫生系兼职教授)开展好海外原味课程“自噬的基础与临床”的讲授,利用信息学方面的基础,协助公共卫生系推进线上线下联合教学模式的推广,在适当条件下,可将以上教学模式向公卫基层组织推广;争取开设主讲课程“肿瘤毒理学”一门,讲授肿瘤毒理学机制的最新发现和进展;

2.科研带教方面:抓好研究生及本科生培养,争取在原有的基础上形成一个本科生早期科研训练的特色体系,并配合推动公共卫生系和国际顶尖公卫学院的本科生联合培养工作;

3.学科建设方面:努力履行毒理学科系秘书的任务,协助公共卫生学院及毒理系努力开辟新的研究方向;

4.积极参加学校、学院的社会工作,深化目前已经建立的合作伙伴关系(美国MD Anderson癌症中心,美国NHS癌症中心、浙江大学附属医院等),继续拓展新的学术合作方向,并协助完成本科生及研究生的招生工作。
