缺血性脑损伤是一种高发病率、高致残性、高致死性的疾病,目前除缺血后短时间内进行溶栓外尚无其它很好的治疗手段。申请者近年来在国家自然科学基金面上项目和浙江省科技厅等课题的资助下,针对缺血性脑损伤的药物治疗新靶点和新策略展开系列的研究,主要有以下几方面的成果:

(1)对糖尿病加重缺血性脑损伤的机制与干预进行了系统研究。申请人首先发现annexin A2的糖基化可引起纤溶系统失调,这是糖尿病患者脑梗死后溶栓不敏感的根本原因,而抑制annexin A2的糖基化可有效的改善溶栓效果,这为临床上糖尿病患者的个体化溶栓治疗提供了重要的理论依据。此外,自噬参与了高血糖加重的缺血性脑损伤,而这可能FAK的激活有关。在药物干预方面,首次发现羟基红花黄色素A、异鼠李素、依达拉奉等药物具有很好的抗糖基化作用并能改善糖尿病引起的脑血管损伤,这为其在糖尿病加重的脑缺血损伤中的应用奠定了理论基础,有望进一步对其进行药物的临床转化研究(Stroke, 2012, IF=3D6.018 ; Neurosci Lett, 2012; Biol Pharm Bull, 2012; Thromb Haemost, 2013, IF=3D6.09; PLoS One, 2013, IF=3D4.092; J Neurochem 2015, 2016)。

(2)提出中国人群的缺血性脑损伤后抗血小板二级预防的新策略。脑卒中后目前缺乏很好的治疗方式,目前抗血小板药阿司匹林或氯吡格雷都在指南中作一级推荐,但其在临床不同的人群中疗效、不良反应、依从性的差异目前还不明确。申请者在临床中纳入了1000多例首次的非心源性脑梗死患者,对其进行了5年的随访跟踪,最后得出基于真实世界的研究结果,提出不同的ESRS应该用不同的抗血小板预防,此结果将受益于中国近3千万的脑卒中患者。此外,申请者对当前中国脑卒中的相关危险因素的临床实践进行了调研,从而提出当前在高血压治疗药物的使用、心内膜炎的流行病学与治疗方案与临床指南的差距,进而促进临床诊疗的规范(Transl Stroke Res 2017, 4.502; PLoS One. 2016, 3.057; . Int J Antimicrob Agents 2013,4.415)。

申请人近五年来主持了3项国家自然科学基金面上项目,发表SCI收录论文15篇,影响因子4-5分的3篇、影响因子5-10分的1篇,获得专利1项;培养博士生1名,硕士生2名。

 

三、岗位工作思路&= #21450;预期目标

1)科研:继续目前关于缺血性脑损伤机制与干预的研究,计划从相关基因的组蛋白修饰调控等途径对缺血性脑损伤作系统分析。发表高质量论文(IF>10.0)1-2篇。五年内争取获得国家自然科学基金海外合作项目、国家自然科学基金重点项目等课题等,申请国家发明专利1-2项,促进科研的转化。

2)教学:认真指导研究生进行科学研究;完成基本教学任务,承担药理学教学。承担学生思想政治工作及其他社会工作,立志做一名学生满意的好教师。

3)学科建设:承担学科工作,服从学科安排,增加与同领域科研人员的交流与合作。积极参与国内和国际学术活动并力争在有影响的国际学术组织中任职,为提升学科影响力做出贡献。
