二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

1.  利用小分子化合物组合处理小鼠胎儿成纤维细胞(MEFs),可同时转分化获得来自不同胚层的多种成体细胞。该项工作已于2017年1月在线发表于Cell Research 杂志,本人为第一作者和共同通讯作者。

2.  利用C1-96单细胞mRNA-seq分析平台对血清培养条件下的小鼠胚胎干细胞(mES)进行异质性分析。该项工作已于2016年2月发表于Cell Reports 杂志,本人为共同第一作者。

3.  利用本课题组自主研发的微孔板单细胞分析平台,对小鼠的33个组织器官的细胞进行无偏好性的单细胞分析,建立小鼠单细胞图谱,测序细胞总数高达30万。目前正在进行后期的数据整合工作。本人为本项目的第一完成人。

4.  利用C1-800单细胞mRNA-seq分析平台对人primer胚胎干细胞,人naïve胚胎干细胞以及8d EB 进行单细胞测序分析,目前正在进行功能验证。本人为本项目的第一完成人。

本人2014年12月入职以来,主要的学术成就如下:

1.  利用小分子化合物组合处理小鼠胎儿成纤维细胞(MEFs),可同时转分化获得来自不同胚层的多种成体细胞。利用biomark单细胞qPCR技术对转分化获得的细胞产物进行分析鉴定,包括心肌细胞、平滑肌细胞、星形胶质细胞、神经细胞、脂肪细胞、上皮细胞、肝细胞等,且中间过程不存在被重编程成iPS的阶段。将小分子组合处理第六天的细胞,通过在不同的培养基中继续培养12-16天,可分别高效的转分化成心肌细胞、星形胶质细胞和脂肪细胞,说明小分子组合处理第六天的细胞处于一种具备多向转分化潜能的状态。成体细胞转分化能够获得不同命运的三个胚层的细胞类型,在这一过程中科学家们使用的小分子化合物组合却大同小异。之前的科学研究,科学家们只是关注某一种世系细胞的转分化,转分化产物中的其他副产物大多被忽视。本项目的特色在其所描述的多向转分化过程是细胞命运改变的一种全新的模式。该研究揭示这一全新生物过程中的动态分子调控机制。本项目的研究,不仅能填补人类在细胞命运决定过程中的知识空白,也能够为得到传统方法难以获取的细胞类型提供全新的思路。该项工作已于2017年1月在线发表于Cell Research 杂志,本人为第一作者和共同通讯作者。

2.  利用C1-96单细胞mRNA-seq分析平台对血清培养条件下的小鼠胚胎干细胞(mES)进行异质性分析。小鼠胚胎干细胞的基因表达异质性一直都是一个非常重要的科学问题,然而导致基因表达异质性的机制之前一直研究的都不是很清楚。通过对传统血清培养条件下的小鼠胚胎干细胞进行深度的单细胞mRNA-seq分析,我们发现高差异表达的基因簇和染色质的状态有很大关系,而且二价基因更易于差异表达。同时,我们也揭示了从naïve状态向prime状态转变的最初的pathway。最后我们发现很大一部分细胞内基因表达的异质性都与细胞外的培养条件相关,无血清培养条件能大大的减少细胞的异质性和转录本的异质性。该项工作已于2016年2月发表于Cell Reports 杂志,本人为共同第一作者。

3.  小鼠这一物种是生物基础研究和临床转化研究最重要的哺乳动物模式生物之一。小鼠的每个器官和组织都是由几种到数十种不同的细胞组成的,他们协调作用以维持正常的功能。然而,到目前为止,小鼠做为这样一个最为重要的模式动物,每个组织一共有多少种不同的细胞类型,同一种细胞在不同组织中的基因表达模式的异同等问题都没有得到清晰的解析。利用本课题组自主研发的单细胞微孔板测序平台,对小鼠主要的33种器官无偏好性的进行单细胞转录组建库测序,测序总细胞量超过30万,远远超过目前发表的单细胞测序数据。我们除了对33个器官的细胞进行单个器官的细胞类型组成分析,还对全部数据整合分析,初步定义了88个细胞群类型,接下来我们还将对每个群进行细分,已鉴定不同的细胞种类。此外,我们还分析了几个重要的细胞类型在不同组织中的同一类细胞的异同,如基质细胞、树突状细胞、血管内皮以及巨噬细胞。最后,我们建立目前世界最全的小鼠单细胞图谱数据库,并构建了一个网站,只要使用者输入一个细胞的数据,系统就会自动比对返回该细胞与我们已获得的组织细胞种类的比对结果。本人是该项目的第一完成人。

4.  人多能干细胞具有自我更新和多谱系分化的能力,可以为再生医学和发育学研究提供丰富的材料。但是目前人多能干细胞体外分化仍然面临几个关键的问题,包括分化效率低下,目的细胞功能不完善等。导致这些问题的原因之一是我们对体外建立的分化路径了解的不够全面,传统的实验方法让我们无法系统性地了解分化过程中细胞状态的变化,导致我们无法有目的性地对分化方案进行优化和质控。因此,我们结合单细胞mRNA深度测序技术(C1 HT 800)和拟胚体(EB)分化技术来构建人多能干细胞早期分化图谱。通过对单细胞基因表达数据的分析,我们可以从拟胚体分化过程中发现三胚层(中,内,外)组织细胞的前体,包括神经,上皮,肝脏,心肌,骨骼肌,内皮等。结合多能干细胞的单细胞基因表达数据,我们对各胚层的分化进行了时序性的分析,揭示了从多能干细胞到各胚层早期分化过程中的特异性基因调控模块。同时我们也在单细胞水平上比较了传统的人多能干细胞(prime hES)和幼稚多能干细胞(naive hES)的基因表达谱,发现幼稚多能干细胞在表达谱上更倾向于中内胚层,而传统的多能干细胞具有更多的外胚层基因表达特征,这一点在我们的分化实验中也得到了证明。基于建立的三胚层分化通路的基本框架和对多能干细胞状态的解析,我们可以优化现有的人多能干细胞体外分化系统。本人为该项目的第一完成人。

5.  此外,本人还以课题指导者或者合作者的身份参与了其他课题,如人造血干细胞的扩增,人血液疾病单细胞图谱的构建,人转录组单细胞图谱的构建等。


三、岗位工作思路&= #21450;预期目标

  在科研方面,继续开展单细胞mRNA-seq的工作,对小鼠单细胞图谱的数据进行进一步的挖掘,并进行论文攥写和投稿工作;对人胚胎干细胞分化的课题开展下一步的功能验证实验工作;建立更高效、更低价、更高通量的单细胞测序平台;以第一作者或者通讯作者发表CNS及子刊水平的学术论文2-5篇。

  在教学方面,今年年底将结束入职前三年免教学任务的保护期。我将加入一个教研组,承担本科生和研究生的课程教学工作,教学量至少达到医学院要求的基本时数,积极参与教学改革等工作。

  在研究生指导方面,指导2016级硕士研究生蒋忻怡和2017级硕士研究生周子茗完成人单细胞图谱的构建工作。第一个学生毕业后,将申请博士生导师资格,期待培养更多高质量的研究生。

  积极参与学院、学科、中心等运行管理事务。
